At a Glance
- FDA approved tucatinib with trastuzumab and pertuzumab for maintenance treatment of adults with unresectable locally advanced or metastatic HER2-positive breast cancer.
- The indication applies after induction treatment in patients without disease progression.
- The approval was supported by the phase 3 HER2CLIMB-05 trial.
- Tucatinib was given at 300 mg orally twice daily in the study.
Who was studied?
HER2CLIMB-05 enrolled 654 adults with HER2-positive unresectable locally advanced or metastatic breast cancer, including patients with or without brain metastases, who had no disease progression after 4–8 cycles of induction treatment with trastuzumab, pertuzumab and a taxane.
Why the approval matters
Maintenance strategies aim to extend disease control after successful induction therapy while balancing efficacy and tolerability. The addition of tucatinib offers a new option in this setting under the FDA-approved indication.
India caveat
FDA approval does not automatically establish the same indication, label, reimbursement or availability in India. Indian clinicians should verify CDSCO-approved labeling, local availability and multidisciplinary oncology guidance before applying the indication in practice.
